The questions, one by one
Does having a NIE make me resident in Spain?
No. A NIE identifies you; it does not show that you live here.
No. The NIE is an identification number. It is not a residence permit and does not prove that you live in Spain, and you can hold one all your life without being resident.

What the Ministry itself says
The Ministry of Foreign Affairs' consulate page describes the NIE as the number for foreigners with an economic, professional or social interest in Spain, and warns that although it lets you deal with the Spanish administration, it does not prove residence. Anyone who wants to live here must first check whether a visa is needed.
Why almost every owner has one
Buying, inheriting or selling a home is an economic interest in Spain, and without a NIE no deed is signed. That is why an owner who lives in another country usually has a NIE from the purchase, without ever having been resident.
What does change with residency
Immigration residence is a permit to live here, and tax residence is something else that the tax law decides by calendar year. The NIE decides neither: it neither gives you the first nor binds you to the second.
